BrightLine LED – 优化的荧光滤光片组

过去十年,在荧光成像领域,基于 LED 的光源已经大量取代了气体放电灯(或弧光灯),因为 LED 能够实现更快的通道切换,提供离散的输出波长,消耗更少的能源,并提供显著更长的寿命。在这种激发技术的变化中,需要考虑的一个重要因素是,LED 光源的光谱特征和能量分布不同于弧光灯。

因为传统的滤光片组被设计用于匹配弧光灯的输出光谱,新型 LED 光源的照明光谱和传统滤光片组的透射光谱并不匹配,导致照明设备到达荧光染料的亮度减少。例如,用于传统的弧光灯光源的 DA/FI/TR/Cy5-4X-B 四带滤光片组。此滤光片组中的激发滤光片针对氙气(Xe)或水银(Hg)弧光灯的输出光谱进行了优化。当光源改为亮度近似的 LED 光源时,四个透过带的整体透射能量降低了,激发 Cy5 波段的强度降低尤为显著。

BrightLine 基于 LED 光源的滤光片组的解决方案

为解决频谱不匹配造成的透射能量损失,Semrock 研发了新一代滤光片组,将激发滤光片与当今市场上常用的基于 LED 的光源的独特光谱峰匹配。下面以前面提到的四带通滤光片组的 Cy5 通道来举例说明。图 2 (下图) 显示了在 LED 照明光源中,所选的激发滤光片从 FF01-650/13(传统的 Cy5 激发片)改变为 FF01-635/18 时,传输能量得到了明显的增强。对比 FF01-635/18(LED Cy5 激发片)可看出,FF01-635/18 的峰值已超过 LED 峰值。

使用了新设计的激发滤光片的 LED 优化滤光片组,可以看到,改进后的 DAPI 和 TRITC 波段的传输信号强度比传统的滤光片组增加了 35%。重要的是,Cy5 波段内的信号强度提升了 750%。

Semrock BrightLine 好处

比传统的多波段滤光片组,新一代 LED 优化的 BrightLine 滤光片组提供了几个好处:
