使用纳米晶体的荧光成像

纳米晶体是一种荧光染料,它们吸收光子,然后几乎立即重新发射出更长波长的光子。和传统的荧光染料(包括染料和荧光蛋白)相比,纳米晶体由 Invitrogen Molecular Probes® 公司制造,Qdot® 有一些重要的区别。纳米晶体是纳米级的半导体原子簇,通常带有一个额外的半导体壳层,然后是一个聚合物涂层,该聚合物涂层可以与蛋白质、寡核苷酸、小分子等生物分子偶联,生物分子直接结合纳米晶体和感兴趣的目标。

纳米晶体非常明亮,具有高灵敏度、很弱的标记干扰、以及长期的光稳定性,是某些应用的理想之选:如活细胞成像和动态研究。出色的光稳定性也意味着它们可固定和存档,用于样品的存储,尤其适用于很多应用。因为纳米晶体的尺寸和发射荧光的波长之间存在着直接的关系,因此可以制作出一系列的纳米晶体,每个都有一个窄的、独特的发射光谱,而且全部都由单一的蓝色或紫外波长激发。因此,纳米晶体是高密度多重染色的理想选择。当然,纳米晶体也有其固有的其他特点,可能会限制其在某些方面的应用,例如其较大的物理尺寸和较长的寿命。

为充分利用纳米晶体的优点,对其配套的滤光片产品进行优化就非常重要。Semrock 提供 BrightLine™ 滤光片组,专为广受欢迎的纳米晶体成像应用而设计。一个通用型的长波通发射滤光片可以实现多个纳米晶体的同时成像,亦可使用眼睛观察或彩色相机成像。此外,我们也为单个纳米晶体设计了对应滤光片组。重要的是,这些滤光片组,都属于 BrightLine 滤光片组,都拥有出色的 “不易烧融” 的牢靠性,可很好地匹配光稳定的纳米晶体!
