使用VersaChrome 滤光片进行光谱成像

传统的光谱成像系统通常无法提供薄膜干涉滤光片的主要优点,即高透过率、陡的光谱边缘和高的通带外阻挡。现在有了 VersaChrome 滤光片,这些优势可以在简单的光谱成像系统中实现,应用范围从荧光显微镜到高光谱成像。

在荧光显微镜中演示光谱成像,使用 Semrock VersaChrome 可调式滤光片(TBP01-617/14)将三个光谱重叠的荧光团(荧光蛋白)标记在一个标准正置显微镜的发射通道中,获得了一个“波长堆栈”图像(对应于一系列几乎连续的发射波长)。图1显示了以1nm 间隔拍摄的 61 幅图像中的 6 幅,图 2 显示了从仅存在单个荧光团的部分图像中获取的测量强度光谱。用 SYTOX® 橙色标记的细胞核很容易与其他细胞结构区分开来(图1)。然而,由于F-肌动蛋白和线粒体被高度重叠的荧光团标记(Alexa Fluor™ 568 和 MitoTracker® Red,),线性分解是识别相应细胞成分的必要条件。图3显示了用线性分解分解的图像。

值得注意的是,这些可调谐滤光片的光谱特性对于光的s-和p-偏振几乎是相同的,这是使用液晶可调谐滤光片和声光可调谐滤光片不容易获得的特性。偏振独立性对于光谱成像系统来说是有益的,然而在大多数仪器中,电子可调谐滤光片的偏振限制导致了至少一半的信号损失。因此,VersaChrome 滤光片不仅提高了光谱成像的通光量,而且简化了仪器的复杂性。
