多带滤光片组配置

在单个样品中标记多个不同的感兴趣对象的能力大大增强了荧光成像的力量。实现此类样品的高质量成像的一种方法是,在多次拍照之间切换多个单带滤光片镜盒,然后以电子方式组合这些照片。这种方法的局限性是:不同时间点上会存在多个单色图像之间的 “像素位移”,以及速度慢(相对于每次采集一张完整的彩色图像)。

Semrock 使用 Bright Line“- ZERO ”工艺解决了 “像素位移” 的问题。不可否认,单波段滤光片镜盒的方法仍然是很好的技术之一,可以实现图像的高信号噪声比值和减少串色的可能。但随着高速成像需求的不断增加,尤其是对使用荧光蛋白标记的活细胞进行实时成像分析时,需要一种可以替代单带滤光片组镜盒的方法,但是又不要减少太多的图像真实度。Semrock 的多波段光学滤光片产品将多色同时成像提升到一个新的水平。

有三种类型的多带滤光片组可实现同时多色成像。

“全多带”配置

该配置的激发片,发射片和二向色镜分束镜都使用多波段滤光片,是理想的直接可视化的解决方案,适合的应用有:在样本上定位感兴趣的区域。这种方法速度快且容易实现,并与所有标准的荧光显微镜兼容。然而,它需要一个电子成像的彩色相机,不能消除荧光串扰。

“Pinkel”配置

该配置使用多波段发射片和多波段的二向色镜分束镜配合多个单波段激发片。它提供了一个经济的方式来实现非常高速、高信号噪声比值、同时的多色成像。这种方法基于单色 CCD 相机。相对于彩色相机,单色 CCD 的成本较低,提供了更好的分辨率和更低噪声,带来更好的性能。虽然相对于“全多带”的方法,串扰有所减少,但由于所有发射波段同时成像,仍然可能存在的串扰。

“Sedat”配置

该配置使用装配在同步滤光片转轮中的多个单波段激发片、多个单波段发射片,和一个具有多边缘的二向色镜分光镜。该方法为高速同时彩色成像提供了很好的图像真实度,但需要在系统硬件上加大投入。

Semrock 可同时提供“全多带”四波段滤光片组和独特的四色“Pinkel” 和“Sedat”组。
