荧光滤光片介绍

一个典型的系统有三个基本的滤光片组成:激发滤光片(或吸收滤光片),二向色镜分束镜(或二向分色镜)和发射滤光片(或阻挡滤光片)。激发滤光片通常是一个带通滤光片,它只透过荧光染料的吸收波长,从而减少荧光的其他来源的激发光和阻挡荧光发射带内的激发光。二向色镜分束镜是一种边缘滤光片,用于斜角入射 (通常是 45 °),以有效地反射激发波段的光和透射发射波段的光。Semrock 发射滤光片通常是一个带通滤光片,仅透过荧光染料的发射波长,并阻挡这个波段之外的所有不需要的光 – 尤其是激发光。通过滤光片阻挡不需要的激发能量(包括紫外和红外)或样品和系统的自发荧光,就可以得到具有黑暗背景的荧光图像。

Semrock 滤光片如何优化您的荧光应用?

大多数的 Semrock 滤光片组综合考虑了亮度和信号噪声比的要求。这些滤光片组是标准成像条件下滤光片组的好的选择。但是,当样本的信号较弱时,更宽的激发滤光片和发射滤光片的组合可以增加信号采集效率。在单分子成像等研究中,通常使用具有宽带通或长通的发射滤光片的滤光片组。在使用这些滤光片组的研究中,需要通过合适的样品制备流程来保持非常低的背景自发荧光,然而,由于这样的宽带通滤光片组占用了很大的光谱带宽,不适合用于需要多种荧光染料成像的多色成像。

对于多种荧光染料标记的样品成像来说,应该选择更窄的带通滤光片组。这样的滤光片组能显著减少多个荧光染料之间的串色。狭窄的通带只允许透过荧光发射光谱中强的部分。对于具有自发荧光的背景噪声高的样品,可以显著降低样品的自发荧光,从而提高信噪比。高信号噪声比的滤光片组适合用于荧光信号强的样品。
