Semrock 开发了一项新的滤光片工艺:可调式薄膜滤光片,通过调节入射角,在很宽的波长范围内,中间波长 CWL 可连续可调,而光谱性能基本上没有变化。

这项工艺可以为可调式激发端光源增加灵活度。比如,可用于 Fura-2 测钙的实验配置。

示踪钙离子 Ca2+ 浓度变化

荧光染料 Fura-2 的吸收光谱随其附近的钙离子(Ca2+ )浓度而显著变化。我们可使用两种不同激发波长拍摄样品的数字图像,通过测量图像的强度比,就可追踪当样品的功能改变时,所在位置的Ca2+ 浓度变化。

使用 Fura-2 进行比例成像的常规方法是在一个滤光片转轮中切换两个激发滤光片,并使用固定位置的不变的二向色镜分束镜和发射滤光片,通过软件采集不同激发波长的两组时间序列图像,然后通过软件分析、校准、测量,从而得到特定区域(AOI)的钙离子浓度的变化情况。

使用 VersaChrome 可调式带通滤光片,通过旋转切换激发波长,可以得到更高的速度,Sutter Instruments 公司 VF-5 已经将此商业化。
